4D4H - Innovating across four dimensions (Discovery, Design, and Delivery of Drugs) for human health

Programme

Mission and Scope

Demand for more effective, targeted, and safer therapies has never been greater than today in modern medicine. The Innovating across four dimensions (Discovery, Design, and Delivery of Drugs) for human health (4D4H) programme responds to this need by integrating cutting-edge approaches in medicinal chemistry, pharmaceutical sciences, bioanalysis, nanotechnology, drug delivery, and biointerface science. The programme brings together complementary expertise spanning these disciplines, covering the full spectrum from target and biomarker discovery to molecular drug design and synthesis, quantitative bioanalysis, biopharmaceutical development, and advanced drug delivery systems. By combining experimental and computational approaches, 4D4H enables data-driven and mechanistic understanding of drug action, bridging fundamental science with clinical practice. This integrated approach supports the development of smarter, safer, and more effective precision therapeutics for urgent medical and societal challenges.

Research Aims

The 4D4H programme integrates complementary expertise across chemistry, biomaterials, biology, engineering, and medicine to advance precision therapeutics through four interconnected research aims:

Molecular Discovery & Design
Discover disease-relevant targets and biomarkers and design innovative pharmacologic modalities, such as classical small molecules, photoswitchable molecules, biologics, and gene therapies, using medicinal chemistry, biocatalysis, computational modelling (computer- and/or artificial intelligence-assisted drug design), and photopharmacology

  • Drug Delivery & Biopharmaceutical Innovation: Develop advanced delivery systems, hydrogels, and bioengineering approaches for targeted and controlled therapeutic intervention
  • Integrated Drug Development & Evaluation: Create predictive models and apply quantitative bioanalysis (pharmacokinetics and pharmacodynamics, toxicity) to understand and optimize drug efficacy and safety
  • Disease-Focused Precision Therapeutics: Translate these approaches to major disease areas (e.g., cancer, COPD, fibrotic and neurodegenerative diseases, infectious diseases) to enable personalized treatment strategies

Together, these aims span the full 4D4H spectrum: Discovery, Design, and Delivery of drugs for Human Health.

Relevance and Key Activities

From discovery to delivery - Designing the future of precision therapeutics

By integrating complementary expertise across disciplines, 4D4H establishes a connected early drug development ecosystem, linking molecular discovery, drug design, quantitative evaluation, and advanced delivery strategies. This approach enables both scientific breakthroughs and tangible societal and economic impact.
4D4H addresses major challenges in modern medicine by developing targeted, effective, and safer therapies for complex and age-related diseases such as cancer, COPD, fibrotic and neurodegenerative disorders, and infections, aligned with the UMCG focus on healthy ageing.
Beyond conventional approaches, 4D4H combines chemical, biological, and engineering strategies to not only design and deliver drugs, but also to understand, control, and optimize their behaviour in biological systems. This includes emerging concepts such as spatiotemporal control of drug activity, advanced biomaterials, and data-driven therapeutic design.

Through collaborative partnerships, spanning academic networks within the PRECISION Institute, clinical stakeholders at the UMCG, and industrial partners, we have already achieved concrete advances, including:

  • Novel inhalation devices and vaccine delivery strategies;
  • Next-generation gene doping detection tools;
  • Supporting personalized, clinical decision-making in cancer therapy;
  • Reductions in the use of animal models in drug safety and disease research;
  • Better integration of implants with surrounding tissues;
  • Novel nanomedicines to bypass biological barriers

Key Activities

  • Molecular design & discovery: Designing and optimizing therapeutic molecules using medicinal chemistry, biocatalysis, computational modelling, and innovative approaches such as photopharmacology
  • Advanced bioanalysis & biomarker research: Developing multi-omics and MS-based platforms to study drug mechanisms, identify biomarkers, and enable patient stratification
  • Predictive models & evaluation: Establishing quantitative workflows and advanced in vitro/ex vivo systems (3D models, organ-on-chip) to assess drug efficacy and safety
  • Drug delivery & biointerfaces: Engineering biomaterials, nanomedicine, and delivery systems to achieve targeted and controlled therapeutic/regenerative intervention
  • Integration, collaboration & acceleration: Connecting in silico, experimental, and translational research through RUG–UMCG collaboration, supported by automation, miniaturization, and public–private partnerships

Together, these activities create a continuous feedback loop between discovery, design, development, and delivery, enabling more precise and effective therapies.
